(2) The detoxification effect of vitamin C on zearalenone toxicity in piglets.[TOP]

Zearalenone (ZEN), one of the more virulent mycotoxins occurred in various cereals and feed during recent decades and made serious health hazards to plants, animals and humans. Vitamin C (Vc) has been shown to be an effective antidote to zearalenone. In this paper, the effects of diets containing zearalenone on the growth performance, genital organ and immunoglobulin of weaning piglets and the toxicity alleviation of vitamin C were studied. Piglets were weaned at 21 days of age and 32 healthy female hybrid weaning piglets (Duroc × Landrace × Large white) with a mean weight of 12.27 ± 0.30 kg were randomly selected. The thirty-two female weaning piglets were divided into four treatment groups according to body weight: control; basal diet + vitamin C (150 mg/kg); basal diet + 1.0 mg/kg ZEN; basal diet + 1 mg/kg ZEN+vitamin C (150 mg/kg). There were eight replicates in each group. The test period was twenty-eight days. The results demonstrated that dietary zearalenone could significantly increase the length, width and area of vulva (P < 0.05), the genital organ coefficient (P < 0.05), the level of IgA, IgG and IgM (P < 0.05), the level of BUN, CRE, AST and TBIL (P < 0.05), and significantly decrease the level of E2, PROG, LH and FSH (P < 0.05). However, the addition of 150 mg/kg vitamin C to dietary zearalenone prevented deformities in the vulva, decrease in immune response capacity, changes in serum biochemical indicators and disorders in hormones level of the piglets that received the diet containing only zearalenone. In conclusion, feeding ZEN of 1.0 mg/kg can result in a deleterious effect on piglets, which was totally or partly ameliorated by dietary supplementation of vitamin C at concentrations about 150 mg/kg diet. This study systematically investigated the inhibition mechanism of vitamin C on ZEN-induced reproductive toxicity, immunotoxicity and hematological toxicity of piglets, and which provided new ideas for reducing the harm of mycotoxins to the animals through means of nutrition regulation.

(3) HSCCC Separation of the Two Iridoid Glycosides and Three Phenolic Compounds from Veronica ciliata and Their in Vitro Antioxidant and Anti-Hepatocarcinoma Activities.[TOP]

Five main compounds, including two iridoid glycosides (catalposide, verproside) and three phenolic compounds (luteolin, 4-hydroxy benzoic acid, 3,4-dihydroxy benzoic acid), were separated and prepared from the crude extract of Veronica ciliata by high-speed countercurrent chromatography. n-Hexane/n-butanol/water (1.5:5:5, v/v/v) was used for the separation of catalposide and verproside. n-Hexane/n-butanol/water (3:2:5, v/v/v) was used for the separation of luteolin, 4-hydroxy benzoic acid and 3,4-dihydroxy benzoic acid. The head-to-tail elution mode was used with a flow rate of 5.0 mL/min and a rotary speed of 800 rpm. Finally, a total of 1.28 mg luteolin, 6 mg 4-hydroxy benzoic acid, 2 mg 3,4-dihydroxy benzoic acid, 2 mg verproside and 10 mg catalposide with purities of 98%, 99.1%, 99.5%, 99.8% and 99%, respectively, were obtained from 200 mg of crude extract. In addition, their structure was identified using MS, ¹H-NMR and (13)C-NMR. To the best of our knowledge, this is the first report of the separation and purification of iridoid glycosides and phenolic compounds from V. ciliata by high-speed countercurrent chromatography (HSCCC). Among these compounds, luteolin, 4-hydroxy benzoic acid and 3,4-dihydroxy benzoic acid were separated from V. ciliata Fisch. for the first time. The results of the antioxidant activity show that protocatechuic acid and luteolin have strong antioxidant activity compared to 2,6-di-tert-butyl-4-methylphenol (BHT) and vitamin C (Vc). Five compounds also exhibited strong anti-hepatocarcinoma activities.

(4) Attenuation of Oxidative Stress of Erythrocytes by Plant-Derived Flavonoids, Orientin and Luteolin.[TOP]

Erythrocytes are easy to be injured by oxidative stress in their lifespan. Although there are several chemicals such as vitamin C (VC) that would be able to reduce oxidative stress, natural herbal products still remain an interesting research area. The current study investigated the effects of two plant-derived flavonoids, orientin and luteolin, on erythrocytes and their possible mechanisms. This experiment was divided into nine groups, which were normal group, model group, VC control group, and treated groups with different doses of orientin and luteolin (10, 20, and 40 μg/mL), respectively. Hemolysis rate was determined by spectrophotometry. Antioxidative enzyme and products were evaluated by different methods. Erythrocyte cell surface and cellular structure were observed with scanning or transmission electron microscope, respectively. Oxidative stress induced significant increase in hemolysis rate of erythrocytes. Orientin or luteolin ameliorated hemolysis of erythrocytes in oxidative stress in a dose-dependent manner. Both orientin and luteolin reduced oxidative products and increased antioxidative enzyme activities. Moreover, orientin and luteolin attenuated oxidative stress induced damage of erythrocyte cell surface morphology and cellular structure. In conclusion, orientin and luteolin could protect human erythrocytes from oxidative damage by attenuating oxidative stress, protecting antioxidative enzyme activities, and preserving integrity of erythrocyte structure.

(6) Attenuation of oxidative stress of erythrocytes by the plant-derived flavonoids vitexin and apigenin.[TOP]

Vitexin belongs to the flavonoid carbon glycosides, apigenin is its aglycone. Studies have shown that both of them have antiviral, antioxidant and anticancer effects in vivo and in vitro, but the protective effects on human erythrocytes have not been reported. We studied and compared the protective effects of vitexin and apigenin against H2O2-induced oxidative damage in human erythrocytes and explored the structure-activity relationships. The experiment established the H2O2-induced oxidative damage model, with vitamin C (VC) as a positive comparison, to observe the changes of sulfhydryl content in the red cell membrane protein, erythrocyte morphology and membrane skeleton ultrastructure after oxidative damage. Research results showed that vitexin and apigenin could significantly reduce the degree of hemolysis, lower MDA content, and enhance the activities of erythrocyte antioxidant enzymes. In addition, vitexin and apigenin could restore cell surface morphology through protecting the sulfhydryl content in the red cell membrane protein. Among them, antioxidant capacities of apigenin at doses of 15 and 30 μg/ml were better than that of the same doses of vitexin, while effects of 60 μg/ml vitexin and 30 μg/ml apigenin were the same as 30 μg/ml VC. In conclusion, both vitexin and apigenin have protective effects against H2O2-induced oxidative damage of erythrocytes, which might be achieved through directing subdue oxygen free radical and protecting the antioxidant enzyme activity in cells and the sulfhydryl in the red cell membrane protein.

(7) Slight Fermentation with Lactobacillus fermentium Improves the Taste (Sugar:Acid Ratio) of Citrus (Citrus reticulata cv. chachiensis) Juice.[TOP]

The aim of this study was to evaluate the hypothesis that fermentation with Lactobacillus fermentium, which can metabolize citric acid, could be applied in improving the taste (sugar:acid ratio) of citrus juice. During fermentation, the strain of L. fermentium can preferentially utilize citric acid of citrus (Citrus reticulata cv. Chachiensis) juice to support the growth without the consumption of sugar. After 6 h of fermentation with L. fermentium at 30 °C, the sugar:acid ratio of citrus juice increased to 22:1 from 12:1, which resulted in that the hedonic scores of sweetness, acidity and overall acceptability of fermented-pasteurized citrus juice were higher than the unfermented-pasteurized citrus juice. Compared with unfermented-pasteurized citrus juice, the ORAC value and total amino acid showed a reduction, and no significant change (P > 0.05) in the L*, a*, b*, total soluble phenolics and ascorbic acid (Vc) content in the fermented-pasteurized citrus juice was observed as compared with unfermented-pasteurized citrus juice. Hence, slight fermentation with L. fermentium can be used for improving the taste (sugar:acid ratio) of citrus juice with the well retaining of quality.

(8) Optimized Extraction of Polysaccharides from Grateloupia livida (Harv.) Yamada and Biological Activities.[TOP]

Polysaccharides from Grateloupia livida (Harv.) Yamada (GL) were extracted by a heating circumfluence method. Single-factor experiments were performed for the three parameters: extraction time (X₁), extraction temperature (X₂) and the ratio of water to raw material (X₃) and their test range. From preliminary experimental results, one type of the response surface methodology, the Box-Behnken design was applied for the optimizing polysaccharide extraction conditions. The experimental data obtained were fitted to a second-order polynomial equation. The optimal conditions were extraction time 5 h, extraction temperature 100 °C and ratio of water to raw material 70 mL/g. Under these conditions, the experimental yield was 39.22% ± 0.09%, which well matched the predicted value (39.25%), with 0.9774 coefficient of determination (R²). GL polysaccharides had scavenging activities for DPPH and hydroxyl radicals in vitro. The scavenging rates for both radicals peaked at 20 mg/mL GL concentration. However, the positive standard, VC (ascorbic acid), possessed stronger antioxidant activities than GL polysaccharides. Furthermore, the anticancer activity of GL polysaccharides on HepG2 cell proliferation increased dose- and time-dependently, but the positive standard, 5-fluorouracil (5-fu) showed more significant anticancer activity in this study. Overall, GL polysaccharides may have potential applications in the medical and food industries.

(9) Synthesis and assessment of the antioxidant and antitumor properties of asymmetric curcumin analogues.[TOP]

In this study, 12 asymmetric curcumin (CUR) analogues and 5 symmetric curcumin derivatives were synthesized, the antioxidant activity of these derivatives were evaluated by radicals 1,1-diphenyl-2-picryl-hydrazyl (DPPH) assay, 2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) assay, ROO (TRAP) assay and O(2-) (NET) assay and anti-proliferative activities of these analogues were assessed against the human hepatoma cell line (SMMC-7721), the human breast cancer cell line (MCF-7) and the human prostate cancer cell lines (PC-3). Most of the asymmetric compounds showed stronger antioxidant activities than Vitamin C (Vc). Curcumin analogues reducing free radicals contain two reaction mechanisms: H-atom and electron transfer mechanisms. Compound 14 showed the most significant antioxidant activity compared with curcumin and other derivatives. Shorted the carbon chain of 14 can reduce the O-H bond dissociation enthalpy (BED) to improve the antioxidant activity. The antioxidant activity of 25 was similar to curcumin. All of the compounds performed better in an anti-proliferate assay than curcumin, especially compound 25, which exhibited the preferential cytotoxic activity against MCF-7 cells(25, IC50 = 9.11 μM, curcumin, IC50 = 70.2 μM). Considering these data, future studies should be performed to assess the therapeutic values of these asymmetric curcumin analogues.

(10) Optimization of extraction conditions of areca seed polyphenols and evaluation of their antioxidant activities.[TOP]

Polyphenols are functional compounds in plants, which possess many bioactivities beneficial for humans. The aim of this study was to establish a highly efficient method for extracting polyphenol compounds from areca seeds and further to identify polyphenols and antioxidant properties of the seeds. A quadratic general rotary unitized design was used to determine the optimal extraction process. The polyphenols were identified using LC-TOF-MS. By comparison with ascorbic acid (Vc), the antioxidant activities of the ethanol extracts were evaluated using three complementary in vitro assays: inhibition of the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical-scavenging activity, hydroxyl radical-scavenging activity, and reducing ability. The two major polyphenols obtained were epicatechin and syringic acid. The ethanol extracts of areca seeds showed significantly greater antioxidant activity (p < 0.05) than Vc using the DPPH and reducing power assay, but lower ability (p < 0.05) using the hydroxyl radical assay. The results indicate that the areca seed is an excellent food material with potential antioxidant properties.
